Abstract: Neue, auf Platin-, Palladium-, Titan- oder Zinn- basierende Metallverbindungen wurden synthetisiert, umfassend charakterisiert und gegenüber einer großen Auswahl an Tumorzelllinien untersucht. Dabei stellte das Erkennen und Verstehen des Wirkungsmechanismus dieser Metallverbindungen einen Grundpfeiler für die Forschung und Entwicklung neuer hocheffektiver Therapeutika dar. Biokompatible Materialienwie Liposomen und mesoporöses Siliziumdioxid (MS),als Speicher- und Transportsysteme für diese zytostatischen Metallverbindungen, erwiesen sich als vielversprechende Alternative zur gegenwärtigen Behandlung verschiedenster Tumorerkrankungen. Weiterhin eröffnet diese Arbeit durch die gezielte Verwendung von MSals Speichermedium für Metallverbindungen eine neue Perspektive für die Anwendung von Metall-basierten Chemotherapeutika.
NovelPt(II/IV) and Pd(II)coordination and Ti(IV) and Sn(IV) organometallic compounds containing N/O/S/C-donor ligandswere prepared, characterized and investigated among panels of cell lines derived from a broad spectrum of human and animal tumors. Recognising and understanding the in vitromechanism of action of these metallocompounds was essential for development and design of more effective drugs. Biocompatible materials as drug containers and carriers, liposomes and mesoporous silica(MS)surfaces, might be used as alternative to classical approaches, in the treatment of tumors of different origin opening a new perspective for application of metal-based antitumor compounds. In vivo investigations identified a Pt(IV) complex and MS loaded with Sn(IV) compound as high promising in cancer treatment, first by higher reduction of tumor volume compared to cisplatin and second, most importantly, by differentiation of tumor cells into normal phenotype.
