Cette Ligne directrice est conçue pour évaluer des effets, sur l'émergence de plantules et le début de croissance des plantes supérieures, après exposition à la substance d'essai appliquée à la surface du sol ou dans le sol. Des semences sont placées en contact avec le sol traité par la substance d'essai. Les effets sont évalués au bout de 14 à 21 jours après émergence de 50 % des plantules dans le groupe témoin. Les effets observés sont l’évaluation visuelle de l’émergence des plantules, les mesures de biomasse, la hauteur des pousses et les effets nocifs visibles sur différentes parties de la plante. Selon le but de l'étude, l'essai peut être effectué afin de déterminer la courbe dose-réponse, ou, à un(e) taux/concentration unique, comme essai limite. Une analyse statistique appropriée permet de calculer une concentration efficace (CEx) ou le taux d'application efficace (TEx) pour les paramètres d'intérêt les plus sensibles. En outre, la concentration sans effet observé (CSEO) et la concentration minimale avec effet observé (CMEO) peuvent être calculées dans ce test.
