image Systemic and conditional loss of the parkinsonian pyramidal syndrome‐associated protein FBXO 7 ( PARK 15) in mice triggers motor impairment. At the neuronal level, the E3 ligase FBXO 7‐ SCF associates with proteasomes and regulates their assembly. FBXO 7‐ SCF binds to and ubiquitinates the proteasomal subunit PSMA 2. FBXO 7 affects the assembly state and activity of the proteasome in the cells including neurons. Systemic deletion of FBXO 7 leads to premature death and motor disabilities. Deletion of FBXO 7 from forebrain and dopaminergic neurons triggers early‐ and late‐onset motor impairment, respectively.
