This book provides a comprehensive overview of the state-of-the-art methodology to investigate how actin (and actinlike) microfilaments and microtubules work to accomplish different cellular functions in different cellular contexts from bacteria to humans. The book covers extensive and exciting methodologies for protein purification and in vitro reconstitution of several cytoskeleton properties, as well as the functional dissection of microtubule- and actin-based structures, such as centrioles, mitotic spindles, axons, and cytokinetic contractile rings. It also provides a methodology for the study and manipulation of microtubules and actin, including the study of their nucleation, ultrastructural and/or dynamic properties (e.g., using optogenetics), and the functional dissection of post-translational modifications.
