    Abstract: The chemical composition of ether, ethyl acetate and dichloromethane extracts of Umbilicaria crustulosa and U. cylindrica were determined for the first time by HPLC-UV, GC-FID and GC-MS. The HPLC-UV analysis was revealed the presence of methyl orsellinate, lecanoric acid, crustinic acid, gyrophoric acid and atranorin in U. crustulosa extracts while in U. cylindrica extracts were detected following compounds: salazinic acid, norstictic acid, atraric acid, gyrophoric acid and atranorin. GC-FID and GC-MS analysis were enabled the identification of orsellinic acid (40.4%) and orcinol (48.1%) as main components of U. crustulosa extracts and atraric acid (8.5%) and orcinol (10.3%) in U. cylindrica extracts.

    Abstract: Background/Aim. Rheumatoid arthritis (RA) represents an independent risk factor for the development of cardiovascular (CV) disease (CVD). Early detection of atherosclerotic changes is of tremendous importance in the prevention of CV events. An increase in the carotid artery intima-media thickness (cIMT) is considered a sensitive marker of early subclinical atherosclerosis. The aim of our investigation was to assess the cIMT, the number and type of carotid plaques (CPs), and the severity of carotid artery stenosis in RA patients. Furthermore, we investigated the correlation between all the above-mentioned parameters and disease duration and activity. Methods. The research included 92 participants, of which 58 were patients with RA, and the remaining 34 participants were healthy individuals (control group). In patients with RA, clinical examination and laboratory findings were used for assessing disease activity. All participants underwent a color Doppler ultrasound examination of the carotid arteries with a linear probe in order to assess cIMT, the number and type of CPs, as well as the se-verity of stenotic lesions. Results. The mean cIMT in RA patients was statistically significantly higher compared to the control group (0.8 ? 0 .2 mm vs. 0.7 ? 0.2 m m; p 〈 0.01). CPs were found in 34 out of 58 RA patients (58.6%) and 4 out of 34 (11.8%) participants in the control group (p 〈 0.001). The number of CPs per patient was significantly higher in the RA group compared to the control group (1.4 ? 0.9 vs. 0.2 ? 0.4; p 〈 0.001). The cIMT, the presence and number of CPs, and the severity of carotid artery stenosis were not statistically significantly related to disease activity. There was a statistically significant direct correlation between the duration of RA and the percent-age of carotid arterial stenosis (r = 0.320, p = 0.034). Conclusion. The cIMT and the presence and number of CPs per patient were significantly higher in RA patients. More-over, there was a positive correlation between RA disease duration and the severity of carotid artery stenosis. This study showed that RA represents an independent risk factor for an increase in cIMT and the development of sub-clinical atherosclerosis.

    Abstract: Uvod/Cilj. Razvoj tehnike danas omogucio je da se metodom otoakustickih emisija za vrlo kratko vreme dobije uvid u stanje sluha kod deteta neposredno posle rodjenja. Cilj rada bio je da se prikazu prvi rezultati sistematskog neonatalnog ispitivanja sluha metodom otoakustickih emisija u Klinickom centru Kragujevac (Kragujevac, Srbija). Metode. Ova prospektivna studija ispitivanja neonatalne slusne funkcije, zapoceta sistematski krajem 2008. godine u Klinickom centru Kragujevac, obuhvatila je decu rodjenu na vreme i prevremeno rodjenu decu i to 24 h posle rodjenja. Testiranje je vrseno pomocu aparata DPOAEs Interacoustics OtoRead-Screener, kao i retestiranje nakon mesec dana kasnije. Rezultati. Od 01. 01. 2009. do 01. 12. 2010. ovom metodom ispitano 1 994 novorodjencadi, od cega 1 778 rodjenih na vreme i 216 prevremeno rodjenih. Prolaznost na testu bila je veca u grupi dece rodjene na vreme i iznosila je 92,5%, a u grupi prevremeno rodjene dece 55,1%. Odgovori su izostajali obostrano kod veceg broja nedonesene dece u odnosu na decu rodjenu na vreme, kod koje su izostajali jednostrano. Rezultati ponovnog pregleda pokazali su u grupi na vreme rodjenih prolaznost 83,7%, a 61% u grupi nedonesenih. Zakljucak. Prolazno izazvana otoakusticka emisija je efikasan metod za ispitivanje sluha novorodjencadi, posto je neinvazivna, brza i objektivna. Njena korelacija sa cujno izazvanim potencijalima je vrlo visoka, sto potvrdjuje njenu pouzdanost.

    Abstract: The aim of this study is to investigate the possibility of using electrospun polylactide (PLA) fibers as a carrier of the phytoestrogen biochanin A. Polylactide fibers were prepared with different contents of biochanin A by using an electrospinning method at specific process parameters. The obtained electrospun polylactide fibers, as carriers of biochanin A, were characterized by means of different methods. The presented results showed that the mechanical properties of PLA have not changed significantly in the presence of biochanin A. Scanning electron microscopy showed that the fine fiber structure is retained without visible deformations and biochanin A crystals on the surface of the fibres. The analysis by infrared spectroscopy showed that there are no strong interactions between polylactide and biochanin A molecules, which is a good prerequisite for the diffusion release of biochanin A from PLA fibers.The release of biochanin A from PLA fibers in buffer solution pH 7.4 at 37 °C was monitored by applying the HPLC method. The rate and time of the release of biochanin A from PLA fibers is in correlation with the amount of the active ingredient in the matrix of the carrier and follows zero-order kinetics. PLA fibers with biochanin A exhibit concentration-dependent activity on proliferation and migration of L929 fibroblasts in direct culture system in vitro, and proved to be suitable for a potential formulation for use in wound healing.

    Abstract: Background/Aim. Experimental autoimmune encephalomyelitis (EAE) is a model of multiple sclerosis (MS), in which we investigated the neuroprotective effect of agmatine (AGM), known as a primary amine produced via the decarboxylation of L-arginine. Methods. Dark Agouti rats were divided into groups: control (C), Complete Freund`s Adjuvant (CFA), EAE rats decapitated on the 13th day post immunization (dpi) (EAE13) and on the 20th dpi (EAE20), EAE animals given three (EAE+AGM13) and 10 (EAE+AGM20) doses of AGM, and healthy animals ad-ministered three/10 doses of AGM (AGM). Thiobarbituric acid-reacting substances (TBARS), SH groups (SH), total glutathione (GSH), glutathione peroxidase activity (GPx), superoxide dismutase activities (tSOD, MnSOD, CuZ-nSOD) and nitrite/nitrate concentration (NO2+NO3) were assessed in plasma and brain structures [whole encephalitic mass (WEM) and brainstem (BS)]. Results. The obtained results showed that AGM treatment successfully attenuated severe clinical deficits in EAE. Applications of AGM in EAE rats induced normalized TBARS, SH, GSH, GPx and NO in WEM. In BS, AGM expressed less prominent effects, inducing normalized TBARS, GPx and NO, but no effect on SH and GSH. In both brain structures, tSOD activity lowered and normalized at the peak and in the remission phase of the disease, post-AGM treatment. The effect of AGM on the MnSOD in EAE was expressed in WEM/BS only in the remission phase as a reduced activity. Conclusion. Milder clinical form of developed EAE in rats indicates promising therapeutic effect of AGM in MS. The activated antioxidant system and suppressed oxidative/nitrosative stress development may denote a successful blockade of neu-roinflammation initiated by EAE immunization. The study implies the capability of AGM to attenuate oxidative/nitrosative damage at the peak of EAE by modulating antioxidative defense capacity during the time-course of the disease. Thus, AGM may be considered as an agent with a beneficial effect on neuroinflammation in EAE.

    Abstract: The essence of rheumatoid arthritis (RA) pathogenesis is inflammation, modification of immune system and cell damage. 5'-nucleotidase (5'-NT) and adenosine deaminase (ADA) have a significant role in the process of inflammation-caused tissue damage. The aim of the paper is to define 5'-nucleotidase and adenosine deaminase activity in serum of patients with rheumatoid arthritis treated with Methotrexate and patients with rheumatoid arthritis who have not been treated with Methotrexate, as well as to determine correlation between the enzymes and disease activity. The research included 160 patients suffering from rheumatoid arthritis, 60 of which have not been treated with Methotrexate (age 56,8; 68,3% female) and 100 patients treated with Methotrexate (age 59,8; 88% female), as well as 60 healthy controls (age 58,8; 66,6% female). Patients suffering from chronic inflammatory diseases, chronic respiratory, cardiac and kidney insufficiency, severe acute diseases and other diseases which might modify inflammatory response were not included in the research. There was no statistically significant difference in 5'-NT values among groups. ADA values were significantly different in all tested groups. Post Hoc analysis (Dunnett T3 test) showed that ADA activity in RA groups were significantly higher as compared to control group (p〈0.001), and that ADA in RA group with MTX was significantly smaller as compared to RA group without MTX (p〈0.001). There is not significant correlation between the disease activity and activities of tested enzymes. We concluded that adenosine deaminase activity was increased in patients with rheumatoid arthritis, as well as that the application of Methotrexate led to the decrease of this enzyme in the serum of patients with rheumatoid arthritis. Activity of 5'-nucleotidase is not increased in patients with rheumatoid arthritis and did not depend on Methotrexate treatment. Serum adenosine deaminase and 5'-nucleotidase were not good indicators of rheumatoid arthritis activity.

    Abstract: Background/Aim. Although chlorpromazine (CPZ) is an antipsychotic drug widely used in clinical practice for a long time, its mechanism of action has not been entirely defined. An extremely difficult managing of patients acutely poisoned with CPZ is additional reason for detailed studying its toxicity mechanisms. In this clinical study, we investigated whether the oxidative stress (OS) mediates CPZ toxic effects in the exposed patients. Methods. The patients were organized into 3 groups: the T-group - hospitalized patients receiving therapeutic doses of 75-150 mg CPZ/day; the overdosed group, divided into two subgroups: the group M and the group S - mildly (CPZ serum concentration: 0.21 ? 0.05 mg/L) and severely (CPZ serum concentration: 2.66 ? 0.25 mg/L) poisoned patients, respectively, and the group C (control group of healthy volunteers). Oxidative stress parameters [total antioxidative status (TAS) and malondialdehyde (MDA) in plasma)] and superoxide dismutase (SOD) activity in erythrocytes were measured spectrophotometrically, and CPZ concentrations in serum were monitored chromatographically. One set of measurements was performed in the group C and T, whereas two sets of measurements (after 24 hours and 48 hours) were done in the poisoned patients, groups M and S. Results. A decrease of TAS and increase of SOD activity were obtained in both subgroups of the poisoned patients, compared to the controls and the group receiving therapeutic doses of CPZ. A significant increase of MDA was achieved in severely poisoned patients, compared to all other groups. Conclusion. Changed oxidative stress parameters in patients poisoned with chlorpromazine indicate involvement of oxidative stress in the toxicity mechanism(s) of chlorpromazine.
